บทคัดย่อ
Sustainable conservation use of genetic diversity is essential to improve the understanding for species preservation and protection. Advances in molecular biotechnology have generated new opportunities for genetic variation. Molecular genetic fingerprints of eleven Gynura species from Thailand were developed using PCR-based markers. The aim involves elucidating there interspecific genetic diversity important for utilization, management, and conservation. Sixteen of twenty random amplified polymorphic DNA (RAPD), fifteen of twenty inter simple sequence repeats (ISSR), and twenty-eight of thirty sequence-related amplified polymorphism (SRAP) primers produced 262 polymorphic fragments (46, 58, and 158 obtained from RAPD, ISSR and SRAP primers, respectively) were used to define the genetic similarity among the Gynura genotypes by dendrogram and three dimensional scaling. Within-group genetic similarities ranged between 0.305 and 0.971. Dendrogram based on three molecular data using unweighted pair group method with arithmetic mean (UPGMA) was congruent and classified the Gynura species into three major clusters. Clustering was further supported by principle coordinate analysis (PCA). The result agrees very well with the taxonomic characters. High genetic polymorphism documented is significant for conservation and indicate that the conservation of existing natural all Gynura species is necessary by the development of different management practices such as: construction of an in situ conservation area, ex situ conservation of the genus Gynura in Thailand.
